In this case, the effect that applies the highest base power between the effects that modify base power will be applied to that Character.

Can I still play this card and activate its [On Play] effect if this card is the only card in my hand?

No, you cannot. In this case, you will also be unable to DON!! −4, and will be unable to resolve the "Add up to 1 card from the top of your deck to the top of your Life cards. Then, up to 1 of your opponent's Characters' base power becomes 0 during this turn." part of the effect.

If an opponent's Character selected by this [On Play] effect has its base power changed by another effect, such as an effect that sets its base power to 6000, what will that Character's base power be?

In this case, among the effects applied to the same Character that change its base power, the effect that changes its base power to the highest value is applied. For example, if an effect that sets the Character's base power to 0 and an effect that sets its base power to 6000 are both applied to the same Character, its base power will be 6000.
