No, they cannot. In this case, the player who played a Character using a Character's effect is your opponent whose Leader is "OP12-081 Koala".

Can I activate this [Once Per Turn] effect when my opponent plays a Character using a Character card's [Trigger] effect?

No, you cannot. Playing a Character using a [Trigger] effect is not considered playing it using a Character effect.
